With Manning still seeking his first Super Bowl Championship in 2006, the Colts trailed the New England Patriots 21-3 in the AFC Championship Game. Instead of folding up, Manning led the greatest comeback in championship game history as the Colts defeated the Patriots 38-34. Manning would then win his only Super Bowl a few weeks later against the Chicago Bears.

As often as Manning gets criticized for not being clutch enough, he does own the record for most fourth quarter comebacks with 40. His most impressive comebacks were in 2003 against the defending Super Bowl champion Tampa Bay Buccaneers, the 2006 AFC Championship Game and coming back from a 24-0 deficit to defeat San Diego in 2012.
